Atten år etter brødrenes opprinnelige "oppdrag fra Gud", kommer Elwood Blues (Dan Aykroyd) ut fra fengselet og oppdager at mye har forandret seg på den tiden han har vært borte. Partneren hans, Jake, har forsvunnet, bandet hans spiller ikke sammen lenger, og barnehjemmet hvor han vokste opp er blitt ødelagt. Elwood innser raskt at han må ta på seg en helt ny oppgave - samle det gamle bandet med hjelp fra en bartender full av soul (John Goodman), konkurrere ved Queen Moussette's (Erykah Badu) musikkonkurranse "Battle of the Bands" og redde en egenrådig, foreldreløs gutt, Buster.
